Rauwolscine: What It Is and Why We Print the Dose

THE INGREDIENT ALMOST NOBODY EXPLAINS HONESTLY

Listen up. Rauwolscine might be the most hyped and least honestly explained ingredient in the entire pre-workout aisle. Brands bury it inside proprietary blends, sell it with words like "insane intensity," and hope you never ask what the research actually says.

We do it differently. Project M777 carries Alfabin® rauwolscine at exactly 1 mg per scoop, printed on the label where you can read it. This article covers what rauwolscine is, what the science actually shows, where the evidence stops, and why it still earns 1 mg of space in the formula. All of it. The honesty is the point.

WHAT RAUWOLSCINE ACTUALLY IS

Rauwolscine is also called alpha-yohimbine. It is a stereoisomer of yohimbine, meaning the two molecules contain the same atoms arranged in a different three-dimensional shape. Same parts, different geometry, and in pharmacology geometry changes behavior. Rauwolscine and yohimbine bind receptors differently, which is why you cannot treat evidence on one as evidence on the other. Both come from plants in the Rauwolfia family. The Alfabin® in M777 is extracted from Rauwolfia vomitoria root and standardized to 90% rauwolscine. That standardized branded extract is what the "purest form" in this article's title refers to: a disclosed, standardized ingredient, not a mystery powder.

Here is the part the marketing never mentions. Rauwolscine's actual scientific fame comes from the laboratory, not the gym. Since the early 1980s, researchers have used radiolabeled rauwolscine to locate and study alpha-2 receptors in tissue, because it is selective for alpha-2 receptors over other receptor families. That selectivity is what made it a useful lab probe. Selective is not the same as high-affinity, and the numbers in the next section show exactly why that distinction matters. Either way, it became a standard research tool. It did not become a well-studied oral supplement. Those are two very different resumes, and most brands are happy to let you confuse them.

WHAT THE RECEPTOR SCIENCE SHOWS

Rauwolscine is an alpha-2 adrenergic antagonist. Alpha-2 receptors work like a brake on norepinephrine release: when norepinephrine builds up, these receptors signal the nervous system to ease off. Block the brake and, on paper, you get more sympathetic drive, the same signaling family behind alertness and fight-or-flight arousal. That is the mechanistic theory behind putting it in a high-stim formula.

The best binding data comes from a 1998 study that compared rauwolscine and yohimbine across cloned alpha-2 receptor subtypes from several species, published in the European Journal of Pharmacology. The honest reading, and this is where selectivity and affinity separate: both alkaloids showed relatively low affinity for most subtypes tested, with one standout exception. Rauwolscine bound the human alpha-2C receptor with high affinity. So it reliably finds alpha-2 receptors rather than other receptor families, but it does not grip every alpha-2 subtype tightly. That is the real version of the "rauwolscine binds tighter than yohimbine" claim you see repeated online: a subtype-specific finding, not a blanket potency win.

And the honest limit: this is binding affinity measured in cells. It tells you the molecule engages the receptor, which is the reason an ingredient earns a place in a formula at all. It does not tell you the size of the effect when a lifter takes 1 mg before training. We label which is which, because those are two different classes of claim and most brands blur them on purpose.

WHERE THE EVIDENCE STOPS, AND WHY IT IS STILL IN THE FORMULA

Here is the boundary, printed because no other brand will print it. Exactly one published trial has put oral rauwolscine in front of an exercise test, and it did not find a performance benefit. Barnes and colleagues gave twelve healthy men a single dose before three 15-second Wingate sprints (Topics in Exercise Science and Kinesiology, volume 4, issue 1, 2023). Mean power, peak power and fatigue index did not improve. Heart rate and perceived effort did not change either. The one signal they did record was higher post-exercise blood lactate without a matching drop in fatigue.

We are telling you that on the page that sells the ingredient, because you should hear it from us rather than find it yourself. Read it for what it is: one small acute study, twelve men, one dose, one narrow question. It tested whether rauwolscine on its own makes a sprint faster. It did not test focus, appetite, fat loss over weeks, or how the compound behaves inside a full pre-workout matrix alongside caffeine, and nobody has published that work yet. The literature on this molecule is genuinely young, and we will keep reading it. If a trial lands that shows more, we will print that too. Everything else you have read about rauwolscine in a supplement ad is borrowed from receptor pharmacology or from studies on yohimbine, a different molecule.

The closest analogue evidence that exists: a randomized, placebo-controlled crossover trial published in 2022 gave 18 physically active women a single 2.5 mg dose of yohimbine hydrochloride, not rauwolscine, before repeated sprints. Versus placebo, mean power and total work were higher, fatigue index was lower, and heart rate and epinephrine were elevated. Interesting, small, and about the wrong molecule. Because rauwolscine has a different receptor subtype profile, that finding does not transfer. It defines the ceiling of what is known about this alkaloid family, nothing more.

What about rauwolscine itself in people? One modern trial exists, published in 2025 in Skin Pharmacology and Physiology (Meng and colleagues, PubMed ID 40532681), and it says everything about the state of the evidence: researchers delivered rauwolscine through the skin of the forearm in 30 adults as a pharmacological probe to study sweating during exercise in the heat. It did confirm rauwolscine blocks alpha-2 receptors in living human tissue. It was never swallowed, and it measured nothing an athlete cares about. That is the entire human evidence base. When we say this ingredient is included for its mechanism rather than proven outcomes, this is why.

WHY M777 RUNS EXACTLY 1 MG

A 2024 review of yohimbine, the related alkaloid with actual human data, published in Neurology International, lays out the case for respect: the authors call it "deceptively potent," note high person-to-person variability in how much reaches the bloodstream, and point out there are no widely accepted dosing recommendations. Low doses appeared tolerable in the research they reviewed. Large doses have documented serious side effects. That review studied yohimbine, not rauwolscine, but it is the best class-level caution available, and we treat rauwolscine with the same discipline.

That is the whole reason behind the number on the label. One scoop of Project M777 is the serving: 1 mg of Alfabin® rauwolscine alongside roughly 213 mg of total caffeine. Two scoops is a deliberate double dose for experienced, stim-tolerant users only: 2 mg of rauwolscine and 425 mg of total caffeine. The label gates that second scoop on purpose. Start with one. Assess. A compound with high individual variability earns a conservative dose and a clear warning, not a shrug.

Compare that to the industry norm: rauwolscine hidden inside a proprietary blend where you cannot see whether you are getting 0.5 mg or 3 mg.
